crude or extract which is better - (Jul/10/2010 )

hi all :P

i have a new model of inflammatory disease and i was thinking of investigating the effect of withania somnifera roots on it... in which form i should start with... the crude form i.e root powder or extract i.e aquous or ethanolic.... and WHY???

please advice!!!!

out of curiosity, how are you going to use root powder to test? wouldn't you need to dissolve them into something first (i.e water or solvent)??

Hello there :lol:

it depends on the assay you are to use ...
i.e in times we use EtOH extracts the others aqueous extracts while the others
we test different organic solvent extracts.

where as the crude one,, i haven't used any ...

as i mentioned before, it depends on the assay.

search for your protocol, am sure you will find the best mode.

i am planning to give withania somnifera ( Ashwagandha) root powder suspension in water (with 2% gum acacia) orally to my animal model to test its effect (in vivo). All i was asking, was wheather it is beneficial to start with root powder (crude form) on a new animal model of inflammation/ autoimmune disorder or i should use extracts or individual or combined components present in its roots?

if you have no protocol how to isolate the drug, or if there is no reference to previous work, start to test crude extract AND enriched fractions in various concentrations

reason: if the drug is insoluble or hardly soluble in water or ethanol, you may find results with the crude extract...

It depends upon your personal intention- whether you like to translate the herbs into therapeutic application or just simply like to prove that the particular herbs control the inflammation/autoimmunity...!

Again, the next situation is, if you like to publish the work then obviously you need the purest form or extracts of the herbs (to prove that the ‘particular extract’ works again the ‘particular disease’, confirming there is no any cross-reactivity or side effects, etc)...

thanks you all for enlighning me...what was there in my mind was that..we use herbs in crude form for therapeutic purposes (ayurvedic doctors give monoherbal or polyherbal formulations generally in powder form)..in crude form all the active constituents are there which might work synergistically to induce the effect. so, when we are studying the effect in some new disease model..we should start by using crude form. once the herb is proved to be effective against the disease then alcoholic or aqueous extracts can be used...then the active constituents(with the therapeutic effects) can be extracted. :)
