Lentivirus production with LNCX vector - Do I have to use Packaging cell lines? (Jun/09/2010 )

Hi there!

I want to use the LNCX vector to intoduce my gene in some cells. I want to infect them. My question is, I have all the plasmids that I use for lentivirus infection with PLKO.1 plasmids (pspax2 and VSVG); Can I use those plasmids with the LNCX vector, or It is neccesary to use a packaging cell line?

The LNCX vector (Clontech) is a retroviral vector, not a lentiviral vector, so you cannot use psPAX2 and VSVG for package. You need to use retroviral package cell lines such as Ampho293, LNX A.

Thanks. And in the other way aroud? can I use the packaging cell lines with a lentivirus vector?

Nope, lentiviral packaging vectors for lentiviral vectors, retroviral packaging cell lines for retroviral vectors.
