NK cell staining - (May/13/2010 )

i'm going to start some nk experiments soon and i did a few test stains of splenocytes. nk cells should be nk1.1 nkg2d double positive but i only seem to see single positives. anyone have advice?

There is your problem- Balb/c mice don't express NK1.1!!!

Actually, several commonly used laboratory mouse strains such as BALB/c, SJL, AKR, CBA, C3H and A do not express the NK1.1 antigen. For detection of NK cells in these strains the monoclonal antibody DX5 (a pan NK-antibody that recognizes CD49b).
