help preparing broth suspension - (May/11/2010 )

I have never used a nutrient broth (always agar), but i need to use it now. My question is how do i inoculate nutrient broth for incubation...i know i have to make a bacterial suspension in saline first, but how much suspension do i put in how much broth (i want to try to keep every preperation at a similar concentration 0.5 Mcfarland for example).

someone correct me if wrong, but depending on the bacterium and volume of your broth, do you not necessarily need to make a growth culture. Instead you can just use colonies from plates and inoculate a colony into the broth. I tend to keep the inoculator inside a 10ml culture in falcon tube after swabbing a colony from blood agar.

you can simply take a colony from your plate and then inoculate the broth with that.
Just use a inoculating loop or needle for it.

Or a sterile pipette tip. Anything will do as long as it's sterile.
