Custom Made Antibody - Looking for a good company (Apr/30/2010 )

All,
We're having some difficulty in our lab with so called commercial antibodies that are specific to a protein we're interested in looking at. Some of the antibodies look decent in westerns but are non-specific when it comes to IHC (using genomic KO's as our negative control). We are thinking of exploring the generation of custom made antibodies, however we don't have much experience with the various companies that make these. Anyone have any suggestions or experience with such companies? Is it possible to send them tissue from a KO animal to use to test specificity before sending it to use to check? Would companies typically give us a refund if we found the antibody not to be specific in IHC by use of a KO?

are you looking for mono or poly or both?

I guess I don't have a preference as long as it is specific. I suppose monoclonal would be better in that regard, as there are three isoforms of the protein we're interested in and would want to minimize cross-reactivity to the isoforms we're not interested in.
