Is there a difference between PBS-/- and PBS +Mg+Ca when performing an elisa assay?

You could use either, but it is normal to use PBS without Ca/Mg. One exception is if your antibody is dependent on Ca for binding (eg the anti-FLAG monoclonal M1).
