Open Reading Frames - (Apr/21/2010 )

Hello there,

I have some questions concerning ORFs
Start y x z stop

1. an ORF in prokaryotes is characterized by having a start and stop codon in the ends. I have an ORF cloned in a vector in E.coli. Following homologystudies in a database I get the idea that x can be part of the active site of the encoded enzyme.
a. Describe why you would come to this conclusion based on the sequences alone.
b. homologystudies show that ever in y a pointmutation has occurred. From what does this shows and what type of mutations could these be?
c. I want to make a (point)deletionmutation in z. Describe a method with a double PCR.
d. To confirm that z is mutated I will sequence the vector. What primers can I use?
