How to pack the FPLC column - (Mar/05/2010 )

Dear collegues!
I'm just starting to work with Amersham FPLC Akta purifier system, so I would like to ask you such thing: how to pack the FPLC columns correctly? Is it possible to do it manually? I have some empty Amersham FPLC columns provided with the chromatogaphy sytem. Do I need some special device for filling it? Maybe you have some protocols or just can describe the procedure.. I would be very grateful for you help!!!

New columns should be provided with short instructions that usually include information on packing - if they're not, you should find them on GE Lifesciences website (they brought Amersham some time ago). Same goes to new media - they should have instructions that include packing, if not - check on manufacturer website or contact sales representative.

Totally agree with K.B.
Packing your own columns is not a big deal and you don't need anything special other than standard lab equipment. You do however, need to be aware of several things regarding how your column functions and what your media requirements are. It's best to get the instructions for your particular set of components. Once you've read the directions, if you needs some tips and tricks, check back with the forum. Also watch your pressure on the AKTA, don't just set it and forget it before you see if your column matrix can withstand the pressure. Best to keep a close eye on it till you find the right flow rate and start with the low end in the instruction recommendations.
