How to culture cells after sorting? - (Mar/01/2010 )

How are cells cultured after subjecting them to antibodies and cell sorting? How are the bound antibodies removed after sorting? What are the preparation steps after the cells are collected?

The culturing protocol depends on what type of cells you have and what you will use them for after sorting. I don't remove antibodies after sorting my cells and I haven't had any issues. If you provide more details on what you are trying to do, perhaps I can help you more.

Hi, thanks for the reply. I will be using different markers to sort 4T1 mouse breast cancer cell line then culture the cells and propagate them afterwards. Is there any other treatment steps that I need to do after sorting? What do you do in your lab? Are the bound antibodies causing any problems?

I just sort into media + FBS + pen-strep and then plate the cells out in whatever media they need to grow. The antibodies are never an issue, and I'm pretty sure they will dissociate pretty quickly at 37 degrees in the incubator.
