transfecting human mesynchemal stem cells - (Feb/09/2010 )

Any idea what's the best way to transfect human mesynchemal stem cells?

Do you have to transfect them or can you transduce them instead? If you use lentivirus you can get 90%+ transduction efficiency without hurting the cells, in my experience.

We work with non human MSCs and either transfect them by electroporation or nucleofection (amaxa).

I used to do transfection/transduction of human BM MSC.
as the previous guy said Amaxa worked good to me for transient expression of whatever you need.

For viral, I think I used pMSCneo?? from Clonetech
I read somewhere cmv-promoter will not work well with MSC.

There are some reports in the literature that FuGENE HD will give acceptable results in hMSC. Promega recently released this product, and we are currently offering free samples at this URL: http://www.promega.com/applications/genexp...ransfection.htm

It would be very interesting to hear your results if you choose to test this reagent. It may allow you to do what you need without making a big investment in an electroporation set-up.
