dna storage buffer - (Jan/07/2010 )

hello

im doing a research regarding the DNA quality in different storage buffers.. currently i have TE buffer, nuclease free water, and ultrapure water as an options for the research. do u hav any other suggestion that can be used as a storage buffer for DNA (extracted DNA)?

ps: can 70% ethanol and 90% ethanol be used as a DNA storage buffer?

EtOH and Iso P are usually used for precipitation steps. so you cannot use them, DNA or RNA don't dissolve in them.

But I'm not sure about other buffers

You have a couple of waters, why not a couple of buffers? Tris (the T in TE) is a buffer, why not try other buffers with a similar pKa and/or buffering range, like Tricine, HEPES, MOPS, etc?

How will the DNA be isolated? What is the nature of the DNA -- genomic or plasmid? How will you assess the quality of the storage buffer?

i try to look for more info on the buffers that u had suggested. tq..
the dna will be isolated using conventional PCI method. im using nasopharangeal cell line for this study. the quality of the storage buffers and the DNA will be evaluated using uv spec. gel also will be run to determine the quality of DNA after the storage.
