DGGE - Use of clamp or not? (Dec/30/2009 )
Hallo all,
I was wondering if it is really needed to use a rich GC clamp when doing an DGGE ?
Do you use it?
I understand that the clamp will prevent futher movement of the samples, but why should I use it? Just to prevent that the samples will run of the gel in case you need to run it quite long?

DGGE can be run without clamp primers...
but u know DNA runs on the Gradient Denaturant gel...
Double stranded DNA... Continue denaturing as it moves towards more concentration...
if Double stranded DNA convert into single stranded then Single stranded DNA will make secondary structure and Function of DNA will become shape of DNA.... It will become more like SSCP...
Clamp primer inhibits to Melt the total DNA and Keep it into double stranded form....

I think you get better resolution with the clamp.
