Site Directed methylation - (Dec/17/2009 )

Hello friends,
I am new to the area of epigenetics and am planning some experiments related to the same. I am wondering if there are any methods similar to site-directed mutagenesis, so as to perform site directed methylation. Any information is appreciated.

Regards to all

Hi Sam,

we have been grappling with this problem also in the past,

have a look at NEB for Methyltransferases that block restriction sites ie: they recognise a particular restriction site and methylates it.

There are also non-specific methyltransferases including CpG methylase (SssI) and a GpC methylase too I recently found out.

Otherwise there are oligo synthesis companies that could generate an oligo for you with a methylated CpG site.

Good luck
