transformation medium - (Dec/09/2009 )

I want to prepare CCMB 80 buffer which is used in transformation. one of the constituents is Potassium acetate (pH-7.0) but we dont have KAc in lab. Can I use Na acetate instead of K acetate. will it affect the tranformation efficiency?

KOH + glacial acetic acid, adjust pH to 7.0
I wouldn't necessarily be fond of doing this with concentrated solutions, at least not without some good protection.

K2CO3 + acetic acid might be a better choice, liberating CO2.

Its seems a gd idea to use K2CO3 and acetate.
Can u provide me exact details of conc of constituents for both KOH+ Ac, and K2CO3+ galcial Ac.
I will try in both way.
