Cell Invasion Assay - possible to analyze with flow cytometry? (Oct/16/2009 )

Is anyone aware of a kit to evaluate cancer cell invasion, whose results can be analyzed in flow cytometry?

Harvesting the cells for FACS would kind of defeat the purpose of an invasion assay unless you are using one of the chamber inserts with cell permeable membrane - in which case ordinary PI staining would do surely?

not an invasion assay as such, but you could possibly add additional detail to your existing invasion assay. Perhaps cell viability, post-invasion or determine expression of various genes post-invasion? One would have to extract cells from the one side of the membrane though.

do you think is feasible if I do something like this http://www.cellbiolabs.com/userfiles/image...0%20Fig%201.jpg , after I harvest the cells from the lower chamber, and then check the living cells concentration of the FACS ?

well yes but it all depends on what your question is. Are you interested in the cell viability after invasion? Maybe staining for something else may give you better info because i think most cells which are invasive are also viable. But sure, it works and according to the link you supplied, its known.

thanks

do you have any suggestion on wich kit to use or how to build an experiment like this?
