Change in pH after adding Genticin to the media - phenol red changes to yellow (Oct/07/2009 )

Hi,

I am making stock solutions of neomycin ranging from 200ug/mL to 2mg/mL to perform a kill curve experiment. But after adding neomycin to the medium, the color is changing to yellow, particualrly in the ones with higher neomycin concentration. Is this media good for the culture of my 293 cells or should I readjust the pH? if yes, how? Do you encounter the same situation or am i doing it wrong? As you can see I am kind of new to cell culture.

I readjust the pH with 0.2 um filtered 0.1 M NaOH directly to the medium bottle after adding neomycin.

you can also add sodium bicarbonate (NaHCO3) itīs harmful even in high dosis and many media contain it eg DMEM
