Taq Polymerase - Taq Dilution (Oct/01/2009 )

hi folks,

can anyone tell me if to make taq polymerase can be diluted in MilliQ Water to make a dilluted working solution from the original stock?

Probably not a good idea. Most enzymes like a buffered solution containing some salts. If you are going to use the taq immediately after dilution I would suggest you use x1 reaction buffer.

Taq requires low levels of non-ionic detergents for stability. A better solution would be to use Taq storage buffer (20 mM Tris-HCl, pH 8.0 @ 25°C, 100 mM KCl, 0.1 mM Na2EDTA, 1 mM DTT, 0.5% (v/v) Tween 20, 0.5% (v/v) Nonidet-P40 and 50% (v/v) glycerol). An advantage with this is that you can store the dilution for months at -20oC.
