The paper linked below may have some useful information. One potential issue with AK assays is the relatively short duraiton that AK remains active after release from cells. Many cytoxiicty assays require long incubation times, making this diffciult to work around.

Note, the work in the paper I linked was performed by Promega scientists and I work for Promega. It is a peer-reviewed publication and I think this may provide some useful information.
