Buying a new real-time PCR system... - ... Rotor-Gene vs AB 7500 vs ? (Jun/30/2004 )

Hi!

I am in the process of buying a real-time PCR platform. Although fearly new on the market, I though I could fearly trust the ABI 7500. I am planning to use real-time for validation of target genes found by microarray or DD. I like the 96-well plate format. I had lots of problems comparing different plates so I rather have more controls and probes in the same experiment. However, a friend is trying to convince me to give a serious look at Corbett's Rotor-Gene 3000. He says it is more reproducible and allows inter-assay comparison, although I have doubts about it... Also, I am not too hot about having to deals with a rotor type of loading PCR tubes... Is there anybody with experience on either or both machines that could tell me pros and cons about these platforms or any other in a similar price range?

I don't know, if this is what you are looking for, but I used TagMan (Abi7600, I guess) and Light Cycler from Roche, which is also a rotor form, with capillaries. I 100% prefer TaqMan. 96well is fast (esp whan you use the 96 well fast plate - takes 30 min), convenient and you can compare a lot in one run.

If you haven't bought one: that's my 2cent about it, if you have: what did you buy and how is it?

My lab is using ABI 7300 and we love that machine. So I would say go for ABI ones. Oh one more thing, NEVER buy Bio-Rad for real-time PCR machine, its poor performance made my P.I. upset.
