Immunoprecipitation

Immunoprecipitation is a technique that permits the purification of specific proteins for which a antibody has been raised. This primary antibody is either already bound to agarose or can be bound to the protein A/agarose beads during the procedure in order to physically separate the antibody-antigen complex from the remaining sample. Two procedures are described including reagents used.

Immunoprecipitation Protocol
(Abcam)
A general protocol for immunoprecipitation in which an antibody for the protein of interest is incubated with a cell extract so that the antibody will bind the protein in solution. The antibody/antigen complex will then itself be pulled out of the sample using protein A/G-coupled agarose beads. This physically isolates the protein of interest from the rest of the sample. The sample can then be separated by SDS-PAGE for Western blot analysis.

Mammalian Denaturing IP

Non-Denaturing Immunoprecipitation
