Trypan blue will stain dead or dying cells. Viable cells are able to repell the dye and do not stain. Note: Trypan blue has a greater affinity for serum proteins than for cellular protein. If the background is too dark, cells should be pelleted and resuspended in protein-free medium or salt solution prior to counting.

Cell Counts Using a Hemocytometer

The purpose of this procedure is to determine the cell density of the culture. Cell cultures always have some dead cells; the viable and non-viable cells can be distinguished with the use of trypan blue dye and a hemacytometer. Living cells will not take up the dye while dead cells do.
