Desalting resin

we have sephadex g-25, sephadex g-50 and sephadex g-75 resins. our protein is 20 Kda and we would like to desalt it for further processing. Can some one recommend which of these will be suitable. i.e which will cause less dilution and better desalting.

we would be packing them in XK 16/40 column and use for desalting so what amount of sample can be desalted at a time.

bands position in agarose gel

This is a silly question but me not being a molecular biology person am unable to understand this.

we perform plasmid extraction using miniprep kit (alkaline lysis method).

Attached please find the gel. All the samples are of the same clone (lane 5 is hind III marker) but analysis perfomed on different time. Our results should look like the one in lane 1, however we get bands in higher mol wt. Can someone explain this and what we can do to overcome this.
we use TAE buffer during the run and add ethidium bromide during the agarose gel preparation only.

Discard acrylamide solution

HI All,

We have expired acryamide solution 30%. its said to discard acrylamide, polymerise to form gel and discard. can anyone specify the minimum amount of reagents bisacrylamide, APS, TEMED, to add to polymerise this solution so that it will be safe to discard.
