So, my second course Medical Neuroscience started over a month ago, but unfortunately I spent that month in hospital without good internet access. I pretty much regret that, since this course seems to be pretty thorough, with group assigments and other interactive stuff, and planned for and extensive period of 8 weeks. Now I can only watch the videos, without attempting Statement of Accomplishment because some of the weekly quizes are already overdue.
The course stuff said they plan to repeat the course in the future, so maybe then.

You could synthesize it in two parts (with overlaping fragments) in E. coli, or use a modular system of artificially synthetized smaller fragments, that would be complementary (now I just forgot the exact name of this type of cloning stategy, even if you killed me..)
Something like this: A modular assembly cloning technique, for example.
But if the sequence is indeed toxic to E.coli, and you want to use it in E.coli, you will probably face the same problem (but as I understood, you want it for Lactococcus, so maybe this is not an issue).

I was using stock batches of primers many years old.
But also, one time the stock diluted only in water, kept in -20 all the time, thawed only few times.. got completely degraded.
I use only 10mM Tris pH8 to dilute primers since.
