most common amino acid

Hey anybody has some idea about which is the most common amino acid in mammalian proteins? I know it is a very broad question. But still are there any studies like this?

what is 15 O.D.A260? As far as I know about HPLC, we collect fractions based on the retention time, how do collect fractions based on OD?

What calculations do we use for packing any column.i.e. how much volume of beads should be taken for how much compound? For different types of adsorbents, the calculation vary. Like when we run silica gel column we take 1gm slurry for 10mg compound (???)...what about c18, g25 etc.?..I have tried to find out on google for some basic calculation..but did not succeed..
..so experts please help me, I am confused about column chromatography

Confirmation of ligation

After ligation generally we do restriction digestion to confirm if we have got the clones and if the insert is ligated to the vector. But I found on google that many people also do PCR for confirmation. My question is, if the primers spanning to only insert can also be used for this? Whatever colonies we see on plate will be having some resistance, so either it could be ligated plasmid or empty plasmid. But could it be "empty plasmid and insert both in one colony"??
. Then this will also give amplification?
