ofcourse one has to be careful while changing media. Retinoic acid concentrations could also kill at some concentrations, it needs to be titrated to prevent cell death.

I'll try to test on different concentration. Thanks. I've change media every 3 days and wash with PBS. Is it might be due to 'over wash'. I mean that the flow of the PBS from pipette is too strong?

too high concentrations of NGF can kill cells. Try to have a range of concentrations and you also have to change media with NGF every week if you wish to use them for long term.

I try to dissolve NGF with PBS containing 1% BSA but it was not fully dissolve. On the first trial, I treat with 100ng/ml NGF but it's not working. So I increase the concentration for the next trial. The neurite start to grow but on the 5th and 6th day, my cell start to degenerate. Does high concentration of NGF might be the cause of the degeneration?

I have experience with SHSY5Y cells. protocols are quite similar though.

I have not differentiated PC12, only SHSY5Y. The protocols are quite similar.
