Dear Nick, Hello after a long time, good news thst I had a poster presentaion on Nove.13-16.2010 in San Diego, annual neuroscience conference, according thoese results which I got from epigentinc work which i had,(bisulfite treatment) :)

That is not true, you definitely need proteinase K to remove all the protein.
standard methods are just fine for bisulphite conversion

Hi BillyJane, on the contrary you want to proteinaseK digest your genomic DNA to remove proteins that may interfere with the conversion. High quality DNA with a good 260/280 ratio is best for conversion. You must ensure proper denaturation of your DNA prior to conversion, 37C with 3mM NaOH is good, I tend to biol my DNA prior to adding bisulfite. cheers

Hi MethylNick, could you give me a little hint at how to prepare DNA for Bisulfite treatment. I heard recently not to treat DNA with proteinase K cose it fragmentates the DNA..? Thax. BillyJane
