Dear all,
I have MALDI MS results of my 2D gel....Can anyone tell me how to identify the protein using these results. Can any of u give suggestions and software which can help me?

Hi 2 all.....
I did 2D for my control & treated sample for studying the membrane proteins....Manually i could see some spots present in my Treated compared to my control...But i dont know how to interpret my result? We got hoefer 2D view software but i could not succeed in it...Anybody using this software? Can anyone help me out to get report for my data using other means? This work is done for the thesis so kindly help me as soon as possible. Also inorder to identify the proteins i need to pick spots and give for mass spectrometry analysis is enough or I have to give for MALDI also

Dear friends....
I could not succeed in doing mutation....I redesigned the primers and tried from the beginning but i could not get...Im getting colonies after transformation but it is not having plasmid...Please help me regarding this...I have attached the pictures of my results n the protocol i used....In transformation im using controls too... kindly go through n give ur suggestions...

Quantitation of protein concentration in rehydration buffer

I isolated membrane proteins from Thermo Scientific kit...Due to detergents in the reagents I did methanol chloroform precipitation after the isolation and since i have to do 2D...I resuspended the pelleted in rehydration buffer without ampholytes & DTT...
To determine its concentration,what shall i can do without purchasing the 2D quantikit? Is there any other buffer i can dissolve the pellet to quantify its concentration before doing 2D?
Please help me out regarding this?

Hai to all...
I need to run membrane proteins on 2D...I isolated the membrane proteins from Thermo Scientific kit and purified them by methanol chloroform extraction and resuspended in rehydration buffer stock containing Urea,CHAPS and bromophenol blue. From this i will use the working rehydration buffer containing ampholytes and DTT for rehydration overnight. Is this okay?
