I am currently doing a project using the disc diffusion method to test how differing concentrations of antimicrobials affect their bactericidal properties. However, I realise that this disc diffusion method may not be entirely valid as the antibacterials will diffuse at different rates, according to their molecular mass. I have been advised to devise another method to further validate the results and wondered if the phenol coefficient method would be an option.
Does anyone have any recommendations on which is the best alternative method to use?
I'm also testing whether or not different pHs will affect bactericides and have prepared buffer solutions of pH4, pH7 and pH9. What is the best method of applying this - adding the buffer solution to the disinfectant and applying with filter disc to the bacteria as it is inoculated on the medium?
