Please , I want to isolate unknown molecule of lymphocyte from lymphocyte supernatant after incubation for 4 hours which is involved in killing of bacteria how to do that with supernatant
I have a problem with analysis of my result for effect of E.tarda on lymphocytes after separation using percol because i found 3 population on PI graph for + bacteria and just 2 for -ve bacteria
i am preparing lymphocytes for c - culture with bacteria to examine effect of t cell on bacteria but . my cell viability is so low 60 %
procedure :
1- passing head kidney of fish through stainless steel mesh in HBSS
2- centrifugation is 1600 RPm 4C for 5 minutes
3- percol separation
4- MACS separation
