modified boyden chamber (migration assay) for VSMCs

Hey Guys,
I would like to know how much VSMC cells you need in the 24 multiwells insert system, or they call it now the modified boyden chamber. for migration assay

P.S:
I searched online and found that some protocols had used between 30,000 cells - 50, 000 cells. well, I got confued because I use 40,000 cells in 6 wells plate!!! and the 24 wells plate is smaller in size? so howcome?!?!

If anyone had tried it before let me know

Hi everyone :)

I'm performing a migration assay on HCT116 cells, some call it "wound assay"
anyhow, everything is going well with the scraching technique and so.
but when i examine the cells under miscroscope I see lots and lots of floating cells over the scrach and almost all the well.
(picture attached)
Its like the media is not clean enough, hince: I wash with PBS first then add fresh medium!!!
Its been annoying, and im afraid it might affect my results.
Im new to the cultuing techniques and if what I metioned has to do with technical things please let me know
