protein extraction from human tissue (very small) to do ELISA

Could anyone please provide the protocol to extract protein from the tissue that can be quantiifed by ELISA. Please also add the suitable tissue collection method (except snap freeze).

Protein precipitation from RIPA buffer to perform ELISA

Hello,

I have extracted protein from tissue in RIPA buffer.  I have got good yeild but I understood from the Kit protocol/ company's technical support that RIPA buffer cannot be used for doing ELISA's. Could anyone please tell me a working protocol to precipitate protein out of RIPA buffer so that I could use it for ELISA's. .
