Dear sir,
I'm getting problem in protein extraction from human tissue.
I collected sample from OT in ice. In the lab i started to homogenize in glass homogenizer with 10 times volume of Tris Hcl, pH-7.4 and protease cocktail (Sigma). after homogenization resultant pallet was re-suspended in Rehydration buffer. When i took OD, it was almost nil. Than i confirmed through SDS-PAGE. Similarly, got very fainted bands. What may be the reason?
