small dots in cell culture

Hi everyone
I work on PC-3 and NIH-3T3 cell lines. It is for 2 weeks that I have small dots in my culture that slow the rate of cell growth. they appear after the first passage of defreezed cells but in free culture medium they don't appear. Thses dots result in cell death and collapse. has every body any helpful idea?

DNA fragmentation

Hi to All

I am trying to assess apoptosis induction by DNA fragmentation (DNA laddering).
I lysed my cells by lysis buffer and purified DNA by Phenol-chloroform- isoamylalcohol. But unfortunately I have'nt detect the ladder until now. I seed the cells in 6 well duplicate for each concentration. and I run 6microgram DNA in agarose gel .
I would be glad and appreciated  If anybody introduce a sufficient protocol.
