Hi there,
Is there any standard protocol available for the extraction of red blood cells from whole blood?
Thanks

miRNA quantification

There are some commercial miRNA quantification kits available (Qiagen, Exiqon, ABI etc) Are there any specific reasons to prefer one commercial miRNA quantification assay over the other? What are your experiences?
Whole blood sample (ex vivo) stimulation for downstream RNA-assays
03 March 2013 - 12:34 AM
We plan to investigate the influence of growth factors on the bloods' transcriptome. Instead of doing stimulation assays in cell culture, I was faced with the idea to directly stimulate whole blood samples ex vivo (as described for some cytokine assays). The argument is that this is the only way to identify common RNA-signatures influenced by growth factor stimulation.
While I do have my own opinion towards this assay I just want to collect pro's and con's for this procedure.
Best, D.

Hi there,
Does anyone know how long freshly prepared PBMCs can be kept in cell culture?
Thank you

whole blood stimulation for downstream rna-arrays

Hi there,
we plan to investigate the influence of growth factors on the bloods' transcriptome. Instead of doing stimulation assays in cell culture my boss came up with the idea to directly stimulate whole blood samples ex vivo (as described for some cytokine assays). His argument is, that this is the only way to identify common rna-signatures influenced by growth factor stimulation.

I highly appreciate your expert opinion.
