Adipogenic differentiation and senescence marker

Hello, I'm wondering how long I need to differentiate (adipogenic) my cells so that I can do qPCR and Western for senescence markers.  I'm on day 5 of adipogenic differentiation and I was thinking of extracting RNA and protein tomorrow on Day 6.  When are secencent markers (p21 etc.) expressed in differentiating adipocytes? Thanks in advance!

Phosphoryaltion causes inactivation?

Does anyone know why phosphorylation of a protein might downregulate its activity.  It seems that in most cases, like a signal transduction pathway, that phosphorylation activates the protein. Any specific examples? Thanks in advance!

Injecting mice with lentivirus

Hello all,
I am new to lentiviral production and I'm in the middle of writting my proposal.  I need to clone my GOI into a lentivirus, transfect them into 293 cells, test for expression and, then inject them into mice.  I'm studying adipogenesis as it relates to obesity, so can I just inject my clone into subcutaneous fat of mice?  Any help is greatly appreciated!
