Provided the scale bar has a measurement on it, it should be enough. You could if you like also supply the magnification in the text somewhere, but it is not absolutely necessary.

try 8% gel. My proteins are 140 and 160 kDa and it works fine. I usually get beta actin at 8%.
#123035 Protein of 160 kda is present on 6% gel but gapdh housekeeping protein is not.

yes, they can, if you don't over run. it would be better if the gel was 7.5%.
