How to know antibody is ChIP grade?

10 July 2012 - 08:51 AM

Hello,

I have raised a polyclonal antibody against a protein, which is part of a DNA-binding complex.
what procedures should I use to qualify my antibody for ChIP? The binding sites of the complex is unknown. I can do ChIP and reverse the DNA-protein crosslinks and run the eluted protein on Western, but the problem is that there might be free complexes that are not bound to DNA and this experiment will not show that my antibody is a ChIP grade one.
What do you do to show that the antibody works for ChIP?
Thank you,
FHN
