Hi,
I am trying to find a way to fluorescent-label RNA molecules and track them in the cell by microscopy. My idea is to synthesize the RNA (in-vitro transcription) and transfect them into the cell. I know that you can incorporate fluorescent labeled dNTPs into the RNA (like the way one would synthesize RNA-FISH probes), but is the RNA structure/function would be unaltered? Is there any method for adding a fluorescent label to the end of the RNA, without messing with the main portion of it?

ligating blunt ends of a linearized plasmid, is kinase necessary?

A quick cloning question:
I want to delete part of a plasmid. I double digest with to REs which result in sticky ends. I gel-purify the desired linearized fragment. I blunt-end with T4 polymerase and the ligate the ends together by T4 ligase.
Before the ligation, do I need to use T4 kinase to add phosphate groups to 5' ends?

What does "Under Editorial Consideration" mean?

Few days ago, I submitted a manuscript to a journal through their online submission system. Now the tracking system shows the following:
Under Editorial Consideration - Check Status
The manuscript below has entered the review process.
Does this mean that they have already sent it out to the reviewers? Or it just means that an editor is evaluating it?
I think the online submission system is very similar to other journals, so I thought someone might help me to understad this.
