9CFR virus testing (on bovine serum)

Good day,

I want to test our FBS according to "9 CFR 113.47 - Detection of extraneous viruses by the fluorescent antibody technique".

This means I have to test the serum for following viruses:

- Bovine Viral Diarrhoea Virus (BVDV)
- Bovine Reovirus
- Rabies Virus
- Blue Tongue Virus
- Bovine Adenovirus
- Bovine Parvovirus
- Bovine respiratory syncytial virus


I contacted about 50 antibody suppliers, in the hope that anyone would have the whole panel in stock. I thought that would be easy, considering the test is part of a federal regulation. But far from it... Some had maybe 1 or 2 of them in stock, but of the few that actually got back to me none had all of them or neither an idea of where i could get them from. I also contacted the ATCC and they only told me something about proprietary information that they couldn't share... So how are people supposed to adhere to this standard, if no one seems to offer a straight forward kit or panel? I would prefer not to get my antibodies from 5 different suppliers and having to validate them all by myself, but it seems like that is what i will have to do.

So my question is, does anyone here test their biologicals according to 9CFR? And if so, where do you get your antibodies from?

Is this contamination or cell death?

Hi Guys,

I just recently started cell culture and got my first cells in and already something seems fishy.
I have 2 celllines, MRC-5 and BHK-21. I grow both of them in the same medium (DMEM high glucose +10% FBS).
They both grow, but in the BHK-21 flasks I see a lot of cells detaching and swimming in the medium. And after some time these things here show up:

So far I took some of the supernatant and put it on TSA and SAB agar plates and incubated them at 37°. After 5 days in the incubator there is nothing growing on them yet. Can anyone of you more experienced culture users tell me what I am dealing with here?

What is the activity of 1X Trypsin solutions?

Hi there,

when googleing I can either find the activity of 1 mg Trypsin, which seems to be around 150 - 180 units (TAME) which is around 3000 - 3500 USP, or I find things like this: "1X Trypsin-EDTA 1:250, 0.25% (w/v) solution".

How would I calculate the activity of trypsin in a 1X solution?

Different media requirements for multiwell plates vs cell culture flasks?

Hi there,

I am going to grow cells in multiwell plates and harvest them at regular timepoints. I was wondering if the lids of those plates close tight enough to prevent gas exchange with the medium or not, and if the medium therefore needs special additions.
As I just read in a protocol they add NaHCO3 to the medium for the multiwell plates, but not to the medium for the regular cell culture flasks.

How much liquid nitrogen for vapour phase storage?

I have a 22 litre liquid nitrogen storage tank and would like to know how much LN you would put in there to store samples in the vapour phase.
