Fast/easy way to measure cell size

Dear Bioforum members,

I am looking for a technique and a protocol (or just the reference) that will show changes in cell size. I know this could be done using flow cytometry, but am just curious if there are other options out there and whether they are easier and faster to perform. My cells undergo cell cycle arrest upon treatment, so I'll have to control for that too:

Software for scanning of specific aa sequence in proteins

Hello everyone,

I was wondering if there is a software or a database (free preferably) that is able to scan for the presence of specific amino acid sequences/ motifs in proteins. I am interested in finding a phosphodegron motif. It would be cool if the accessibility to this motif could be measured (put it in the context of secondary and tertiary structure), but this feature is not crucial, since I can potentially check it later (it will be more labor-intensive though). Thanks in advance for any comments.

siRNA modifications

Hi,

I would like to perform a knockdown experiment and am about to order some siRNAs. I would really appreciate it if someone could share with me if and which modifications render the siRNA more stable. I have previously ordered siRNA from Qiagen without any modifications and at good storage conditions, these siRNAs were only effective for several weeks. Bottom line is, I would like to compromise between specificity/efficiency and stability/resistance to degredation.
