I want to ask a question about the method for quantification of mtDNA copy numbers in skeletal muscle.

I have read three papers that all used the primer/probe set of one mitochondria gene and one genomic gene with Real-time PCR for mtDNA and genomic DNA copy number measurement.

My questions are: What is the underlying reason by using probes? Is it feasible if I use SYBR green enzyme mix to replace probes for mtDNA cope number quantification, but using primers sequences in those papers if my muscle samples come from their same animal?
Also, is probe expensive compared with SYBR Green enzyme mix kit?

Thank you for any input!
