Hi everybody,

I desperately need help! I have tons of data to analyze and my lab uses DIVA to do it (OMG... is that me or this is the stupidest software out there?). We are using the latest version (what I hate even more than the previous one!).

Is there any free software that you would recommend me? I am looking for a software that would do batch analysis but at the same time would allow me to do adjustments (I run several tissues, and I always have to move the gates) and save them per tube. I know flowjo can do it, but I just cannot convince my boss to buy it.

Also, is there anyway I could apply a gate to all my plots in ANY software? Lets say, I have a plot PE vs APC. Than I want to do FITC vs APC. What I wanted was having the same gate applied to all plots that have APC for one given tube (for example). I don't think I am asking too much! LOL.

Any input is highly appreciated!

Thanks!

yeah, too much to ask from free software. Most use WinMDI but am afraid it will not do what you are looking for.

I just had a free 30-day trial of FlowJo. Maybe that would be long enough for you?

I actually burned my free trial... couldn't finish the analysis as I had to be in the lab 24-7 (wasn't really expecting it). Just went through part of the tutorial.

Did you like the software? Is it really worth buying? I am considering buying myself, will see. I hate my lab has money to spend and won't buy things to make our lives easier...

Thanks a lot

I also just had a free trial with FlowJo.  That's the software I used to use in my old lab, and although I previously was used to CellQuest, I quickly changed my preference to FlowJo pretty, is a great software and I highly recomend it. I think it allows you to do pretty much everything you were asking for on your first post.

As for your free trial running off and you not having time to use it, you should contact them and explain and ask whether they can give you another month for free (mention you are thinking of buying it but not sure because you've been to busy to have a proper test).  They emailed me asking if I had had enough time and if I hadn't they'd tried to get me another 30-day free trial, so its definitely worth trying. Also, you could ask someone else in your lab to get the free trial, it was 2 of us here who got it at the same time.

I'll definitely recomend buying it, but not with your own money. Try to get another free trial, and find some time to use it and maybe some time to show it to your supervisor and convince him/her that is definitely worth buying it.  :P

Good luck!

I just finished a 30 day trial of FCS Express and I thought it was much more user friendly than Flo Jo.  It is also cheaper, as long as you are not a commercial enterprise.  Check it out.

Also, you can do what you are describing in Flo Jo.  If you've drawn a region in an FSC-SSC plot, click in the region and you should get another plot  with only that population.  Then you can change the x and y axis to whatever other parameters you want.  You can also drag quadrants to each tube in the workspace window.  That way you should be able to just click through your tubes.  Let me know if that isn't clear and I'll try to find another way to describe  it.

I just downloaded FlowingSoftware (http://www.flowingso...index.php?id=22) and it's great!  Watch the online orientation video first, that helped me a lot.  Free and plenty of capabilities!

FCS Express is the best one for the off-line data analysis on PC.
If you are MAC user FlowJo is better.
