I am trying to put my promoter and gene fused with one reporter gene dsRED1, one experiment design is to overexpress the gene in the mutant and to see the molecular complementation effect. But I am wondering if the reporter gene will affect the normal function of my gene. Normally, when people fuse the target gene with reporter gene, they will consider if they have to insert the 3 nucleotides (one genetic code, or more ), what kind of nucleotides are better for this purpose? Any suggestion will be very welcome.
