We have been using the first strand kits from Sigma and Gibco. As to protocols, most of these kits come with instructions, and my recommendation would be to follow the protocol the manufacturer gives you! With most of these companies, you can download the instructions off their site.

Has anyone experiance first strand probe synetesis from total RNA using SuperScript II RT from lifetech ?  Basically I need a protocol that worked for you.
