Hi,

I have to make 0.5% crystal violet in 25% methanol.  So, if I understand correct is the following calculation corect for making 10ml total volme:


1) 0.05g crystal violet
2) 2.5ml 25% methanol
3) 7.5ml water.

Or do i have to dissolve the crystal violet in 10ml of 25%methanol?

You need to add 0.05g crystal violet + 2.5ml 100% methanol + 7.5ml water.   This way you'll have 0.5% crytal violet in 25% methanol.  
Your calculations give 0.625% methanol.

Dissolve crystal violet in methanol first, then add DW.
