Hello, I trasnfected siRNA to some activated T cells (activated for 1 week by DC) using Nucleofection. However, the viability is extremely low, just about 1% to 2% of cells remain viable. I also tried to transfect some siRNA by Nucleofection into T cells activated by PMA and ionomycin for 24 hours and the viability is very good, more than 80% to 90% for most of the time. So, is there any methods, tips to enhance the viability of the activated T cells after transfection??

Thank you very much!!!
