Hello everyone!

I am going to try out caspase detection by flow cytometry. So far I've found three different concepts on activated caspase detection:

1) Detection of active caspase-3 with a monoclonal antibody in fixed and permeabilised cells
2) Cell permeable caspase substrate that becomes fluorescent upon cleavage (PhiPhiLux by OncoImmunin)
3) The cell permeable fluorescent caspase inhibitor FLICA

Does anyone has experience with any of these methods? Did it work well for you? Did you co-stain the cells with anything else (e.g. Annexin V, PI/DAPI, TMRE etc.)? Or did you maybe use even another method of caspase detection and can tell me about your experience with it?

Thanks in advance for your answers!
