Hi,
I'm trying to optimise a protocol for differentiating THP-1 monocytes into macrophage-like monocytes.
I've been treating the monocytes with 10nM PMA for 48Hrs and then let them recover for 24Hrs after that. I've managed to show that they are macrophage like by demonstrating increased expression of CD11b, a marker for macrophages, with flow.
However there is no sign of CD68 (internal ab) at all. In fact the results on the histogram/dot plot go backwards/to the left of the isotype control.

If any one can give me any insight at all I would be very grateful!
