Hi! everyone
I'm new in DNA extraction from Cell culture.

I want to extract viral DNA from HSV-1 infected Vero cell

Pls, tell me
-whether phenol-chloroform is good and will get high yeild DNA.
-Do i have to detach infected vero cell by using trypsin before extraction?
-How many MOI that good for infection?
-Do i collect both supernate and cells for extraction?

If you are after pure viral DNA, you will be best to do a virus preparation, and then extract DNA from that. Otherwise, infected cells will have the DNA in them and should be OK to extract with phenol:chloroform straight from the dish.
