sox-2 staining

Hi,

I am trying to staing mouse brain sections for sox-2 (frozen, FIXED WITH PFA 4%).
The problem is that I get a lot of positive stainig - not only in the DG and SVZ, but also all over the brain.
It doesnt look like backgroung. Did anyone tryied to use sox-2 antibody ? do you expect to see so many positive cells?

I have never tried Sox2 on the brain. However, it seems that you are not alone (click here).

I have only done Sox2 in embryonic stem cells, neural stem cells and in E12-E18 rat sections. The staining should be very nuclear and it should only be in areas of neural stem cells (so it should not be everywhere). I an antibody from R and D Systems, Cat # MAB2018 and have never had problems. I have seen Sox2 staining everywhere with other antibodies. GL
